SOT 2024 Poster: Live cell painting - Probing in vitro compound toxicity in high-content imaging

 

This poster focuses on Live Cell Painting for probing in vitro compound toxicity in high-content screens with one of our non-toxic, mix-and-read method with ChromaLIVE. It highlights how it offers the advantage of conducting image-based profiling on live cells, providing new mechanistic insight and overcoming challenges associated with sample alterations in fixed cells.

Applications of ChromaLIVE include : 

  • predicting drug mode of action

  • assessing toxic concentration thresholds

  • evaluating toxic effects indefinitely over time. 

The study validates ChromaLIVE's unprecedented biocompatibility at the genome, proteome, and cell levels, confirming its potential for live-cell assay. It quantifies dose-dependent and time-sensitive cellular events, such as autophagy in live kinetic imaging, enhancing drug toxicity assessment.

A comparison with standard Cell Painting assays on 90 annotated compounds showed ChromaLive yielding the highest mean average precision (mAP) and comparable or better MoA prediction performance.

Furthermore, ChromaLIVE has been successfully used for toxicity assessment in 3D cell cultures, leading to the identification of hit compounds on patient-derived organoids.
